摘要: |
我国四个鲤鱼种群的核糖体DNA内转录间隔区ITS-1进行了PCR扩增、测序.序列分析显示, 370bp的ITS—l序列中GC含量明显高于AT含量, 共检测到34个变异位点, 96个样本得到14个单倍型, 平均单倍型多样性为0.637±0.055, 核苷酸多样性为0.00857±0.00200, 遗传多样性表现较低.黑龙江野鲤种群与建鲤种群问遗传距离最远, 为0.10129, 黑龙江野鲤种群与黄河鲤种群间遗传距离最近, 为0.02305.分子方差分析(AMOVA)表明, 群体间遗传分化系数Fst为0.05373, 几乎所有变异都来自群体内, 群体间遗传分化极小.ITS-1序列构建系统进化树显示, 四个种群分为南北2支, 黑龙江野鲤和黄河鲤种群聚为北方支, 建鲤和荷包红鲤种群聚为南方一支。 |

The internal transcribed spacer 1 (ITS-1) of four common carp populations were amplified and sequenced.The result showed that the average contents of GC were obviously higher than that of AT in the aligned sequences of 370bp.Total 34 variable sites were detected, and 14 haplotypes were recovered.The haplotype diversity and nucleotide diversity were 0.637±0.055 and 0.00857±0.00200, respectively.The genetic distance between Heilongjiang carp and Jian carp displayed the highest, up to 0.10129, while that between Heilongjiang carp and Yellow River carp peculations was the lowest to 0.02305.The fixation indices (Fst) of analysis of molecular variance (AMOVA) among populations was 0.05373, which showed no significant population structure.Tlie phylogenetic tree of ITS-1 built with MEGA 4.1 showed that four common carp populations were clustered into two major clades based on genetic distance.Populations of Heilongjiang carp and Yellow River carp were clustered in northern clade, while Jian carp and Hebao red carp were clustered in southern clade. |
